Molecular Formula | Typical analysis of refined sesame oil shows that it is composed of acids in the form of glycerides, which are: arachidonic acid 0.8%; linoleic acid 40.4%; oleic acid 45.4%; palmitic acid 9.1%; and stearic acid 4.3%. Sesame oil, a complex cyclic e-ther, also contains small amounts of sesamolin (glycoside). Note: Other analytical reports may differ slightly from the data listed above. The description of Sesame Oil in USPNF 20 and the entry for Refined Sesame Oil in PhEur 2002 detail 8 triglycerides in Sesame Oil! |
Category | Carrier for oil-soluble substances; solvent |
Applications | The main use of sesame oil in pharmaceutical preparations is as a solvent for extended-release intramuscular injections of steroidal drugs, e.g., estradiol valerate, hydroxyprogesterone caproate, testo-sterone enanthate, nan-drolone decanoate. Or other oil-soluble drugs, e.g., endocannabinoid decanoate or fluphenazine heptanoate. The elimination half-life of sesame oil from the injection site has been reported to be 23 days after intramuscular or intravenous administration to pigs. Sesame oil can be used in subcutaneous preparations, oral capsules, rectal suppositories, and ophthalmic preparations; it can also be used in suspensions and emulsions. Sesame oil is used as the oil phase in the compounded emulsion formulation of rifampicin, which can be used as a sustained-release system; microemulsions containing sesame oil can be used as a delivery system for ketoprofen. Sesame oil can also be used in the preparation of rubs, pastes, ointments, and soaps. Recently, sesame paste (fahini) containing ground sesame seeds mixed with sesame oil has been investigated as a new type of suspending agent. Sesame oil can be used as an edible oil as well as in the manufacture of margarine. |
Storage conditions | Sesame oil is more stable and less rancid than most other fatty oils; this is due to the presence of antioxidants in its unique composition.PhEur 2001 allows the addition of suitable antioxidants to sesame oil. Sesame oil can be sterilized by aseptic filtration or dry heat. It has been reported that suitable sterilization conditions for injections containing sesame oil are 170°C for 2 hours; 150°C for 1 hour is not suitable(15). However, dry heat sterilization at 150°C for 1 h has been shown to kill Bacillus subtilis spores. Sesame oil should be stored in a full, airtight container protected from light at a temperature not exceeding 40°C. Sesame oil used in the preparation of injections should be stored in a gas-tight glass container filled with inert gas. |
Contraindication | Sesame oil can be saponified by alkaline hydroxides. |
Source | Sesame oil is obtained from the mature seeds of one or more cultivars of sesame, Sesamum indicum Linné (Fam. Pedaliaceae), by pressing in a hydraulic press or by solvent extraction. The resulting crude oil is then refined to obtain sesame oil suitable for food or medicinal use. Further refining improves the odor and color of the oil. |
Management Situation | Included in the FDA Guide to Inactive Ingredients (intramuscular and dermal injections, oral capsules, emulsions, and tablets, and topical preparations). Sesame oil is licensed in the United Kingdom for use in injectable (intramuscular) and gastrointestinal (oral capsules and sprays) preparations. |
