Melanotan II is a lab synthesized peptide hormone that imitates melanocyte-stimulating hormone (a hormone that naturally occurs in the human body). Melanotan II was developed by scientists who were studying the mechanism and the process of signal transduction during the development of skin cancer (melanoma). With an understanding of the melanogenesis process, they developed Melanotan I and later Melanotan II, a polypeptide that would initiate the production of melanin in a natural way hence assisting in the prevention of skin cancer that is caused by UV rays. Alpha-MSH (also referred to as α-MSH or alpha-melanocyte-stimulating hormone) stimulates the melanogenesis; the process by which the skin and hair darkening pigments are produced in mammals. In vitro administration of Melanotan II has been observed to have excellent portent melanotropic action. The structural name of Melanotan II is Ac-Nle-cyclo [Asp-His-D-Phe-Arg-Trp-Lys]-NH2 and its molecular formula is: C50H69N15O9 Melanotan II falls in the class of peptides hormones referred to as Melanocortins.

Product History

The first development of Melanotan II was done by researchers at The University of Arizona, United States. Scientists were searching for a synthetic analog of the human body tanning hormone, α -MSH (Alpha-Melanocyte Stimulating Hormone) as they had already established that melanin was very effective in the prevention of skin cancer (melanoma). From this knowledge, they hypothesized that they could make a UV protective tan by synthetic induction of the body’s pigmentary system. Since the body’s alpha-melanocyte stimulating hormone leads to melanogenesis, melanogenesis initiates the making of melanin (the skins pigment). The scientists went ahead to test if when injected with this synthetic hormone, the body could undergo tanning without exposure to the sun. The results were positive, but there was a challenge, the natural α -MSH had a very short half-life and was therefore not very effective in therapy.

Following repeated synthesis and screening of more molecules in search of an analog that would be longer lasting, the researchers successfully isolated a peptide that was safe and over 1000 times higher in potency compared to the natural α-MSH. This molecule was named afamelanotide commonly referred to as Melanotan I and with more research they developed another analogue which is known as Melanotan II or Melanotan 2. The researchers went ahead to patent and license the two peptides through a technology transfer company to various biotechnology companies that wanted to use them in creating drugs.
