Melanotan produces its photoprotective effects by triggering a 'signaling cascade' via its activation of the MC1R on melanin-producing cells known as melanocytes.
After Melanotan binds to MC1R on the surface of melanocytes in the epidermal layer of the skin, it begins a series of actions and reactions that cause melanocytes to be predisposed to produce eumelanin (photoprotective black/brown pigment).
Initial binding activation of MC1R leads to adenylate cyclase (AC) activation and stimulates the production of cyclic adenosine phosphate (CAMP). CAMP, in turn, activates protein kinase A (PKA), leading to phosphorylation of CAMP response element-binding (CREB). Phosphorylated CREB binds to the CAMP response element (CRE) in the microeye-associated transcription factor (MITF) gene, resulting in the synthesis of MITF protein. MITF can activate multiple genes, including MC1R gene and genes involved in Melanotan production, by binding to multiple genes. This leads to an increase in the concentration of melanase in the melanocytes.
